ABSTRACT

Objective:To understand the status and influencing factors of the bowel function in patients with rectal cancer after sphincter-preserving surgery, and then to improve the patients′ quality of life.Methods:From January 2017 to April 2020, a total of 179 patients with rectal cancer after sphincter-preserving surgery in Sun Yat-sen University Cancer Center were selected as study subjects. General questionnaire and the scale about bowel function of patients with rectal cancer after sphincter-preserving surgery were used to investigate.Results:The total score of bowel function in patients with rectal cancer after sphincter-preserving surgery was (71.82±7.67), and the average score of all the items was (3.99 ± 0.43). In all dimensions of intestinal function, the average score of "defecation affected by diet" was the highest (4.39 ± 0.80), followed by "frequent and urgent defecation" (4.22 ± 0.49), and the average score of "defecation paresthesia" was the lowest (3.03 ± 0.67). Tumor distance from the anal border was the influencing factor of bowel function in patients with rectal cancer after sphincter-preserving surgery ( P<0.05). Conclusions:The bowel function of patients with rectal cancer after sphincter-preserving surgery was well, while the bowel function of patients with rectal cancer after sphincter-preserving surgery with the distance between the tumor and the lower edge of the anus <5.0 cm was relatively poor.

ABSTRACT

Objective:To explore the effects and mechanisms of bortezomib on the proliferation and apoptosis of acute T lymphocyte leukemia cell line Jurkat.Methods:MTT assay was used to test the influence of bortezomib on the proliferation of Jurkat cells.Flow cytometry was used to detect the influence of bortezomib on apoptosis of Jurkat cells.Real-time quantitative polymerase reaction(RT-PCR) was used to detect the effects of bortezomib on the expression of Bax, Bcl-2 and Cox-2 genes in Jurkat cells.Results:The inhibition rates of 5ng/mL, 10ng/mL, 20ng/mL and 40ng/mL bortezomib on Jurkat cells at 24h were (13.23±0.71)%, (39.53±0.95)%, (53.07±1.12)%, (60.43±0.75)%, respectively, and the inhibition rates at 48h were (25.20±0.96)%, (52.80±1.30)%, (60.67±0.64)%, (75.10±1.35)%, respectively.The inhibitory rates of proliferation of Jurkat cells at 72h were (38.37±0.93)%, (60.94±0.85)%, (73.83±5.08)%, (88.37±1.55)%, respectively.The inhibitory rates of proliferation of Jurkat cells increased with the increase of drug concentration and the prolongation of action time, and the differences were statistically significant( F=1 602.202, 1 085.089, 181.034, all P<0.05). Bortezomib (5ng/mL, 10ng/mL, 20ng/mL and 40ng/mL) treatment for 24h, 48h and 72h, the apoptosis rate of Jurkat cells increased with the increase of drug concentration and the prolongation of action time, the differences were statistically significant( F=1 288.571, 223.378, 251.175, all P<0.05). The expression of Bax mRNA in Jurkat cells increased with the increase of drug concentration and time( F=258.446, 518.929, 276.764, all P<0.05). The Bcl-2 mRNA and Cox-2 mRNA expression levels decreased with the increase of drug concentration and the prolongation of action time( FBcl-2 mRNA=236.848, 264.849, 343.968, FCox-2 mRNA=679.404, 1288.681, 1541.850, all P<0.05). Conclusion:Bortezomib can inhibit the proliferation and induce apoptosis of Jurkat cells.Bortezomib can increase the expression of Bax mRNA and decrease the expression of Bcl-2 and Cox-2 mRNA, which may be the molecular mechanism of bortezomib to promote apoptosis.

ABSTRACT

Objective To evaluate the clinical value of tumor chemosensitivity test in the guidance of the advanced epithelial ovarian carcinoma patients treated with chemotherapy.Methods Women with ovarian cancer were enrolled and fresh tissue samples were collected for chemoresponse testing.Oncologists chose a drug for each patient according to the patient’ s condition by double blind method.Each treatment was classified by the assay as: sensitivity (S); moderate sensitivity (I);resistant (R).Progression-free survival (PFS) and overall survival (OS) were detection.The relationship between treatment response and PFS or OS was analysis.Results 262 patients were enrolled.The PFS and OS were significantly improved in sensitivity patients (S) after chemotherapy,but no significant difference were found in I and R groups.In I+R groups (HR=0.66,P=0.008) , median PFS was 8.9 months, OS was 5.8 months.Chemotherapy response test results was consistent with clinical results cisplatin sensitive and resistant ( HR:0.72 vs.0.66).Multi factor regression analysis showed that tumor chemosensitivity test was independent prognostic factors (HR =0.66, P =0.021).The average OS extended for 14 months (37.6 months for the S group vs.group I +R 23.8 months,HR =0.62, P =0.011).Conclusion Tumor chemosensitivity assay is correlated with the clinical effects and might play an important role in guiding the recurrent epithelial ovarian cancer patients with individualized chemotherapy.

ABSTRACT

@#Objective To investigate the reliability of Assessment of Activities of Daily Living of Chinese Quadriplegics. Methods The scale was administered to 20 patients with quadriplegia twice within one week interval by the same rater, and was administered to the same patients by another rater within 2 days after the first assessment. Results The intraclass correlation coefficient (ICC) of the test-retest reliability was 0.994 (P<0.01); the ICC of the inter-observer reliability was 0.971 (P<0.01). Conclusion Assessment of Activities of Daily Living of Chinese Quadriplegics is an instrument with high test-retest reliability and inter-observer reliability.

ABSTRACT

BACKGROUND: The evaluation of stereoacuity can provide an unambiguous assessment of haplopia. It is valuable in assessing the diagnosticaccuracy and the therapeutic effect in strabismus, amblyopia and ametropia. OBJECTIVE: This investigation was designed to evaluate the stereopticdevelopment and the occurrence rate of strabismus in 3-6 years old children, and so that to provide objective data for prevention and treatment of strabismus and steropsis. DESIGN: It was a randomized sampling investigation. SETTING: Department of Ophthalmology,Haikou People's Hospital. PARTICIPANTS:Totally 8326 children in 25 urban and suburban kindergartens, with an age of 3-6 years old, were selected randomly in Haikou city from June 2000 to April 2002. METHODS: 8 326 3-6 years old children in Haikou city underwent vision examinations on hyperopia, eye position and the myopia. Those having eyesight lower than 1.0D or abnormal eye position received atropine drops to dilate the pupil of the eye for optometry. ① 4-6 years old children were examined using standard visual chart and logMAR chart (logarithm of the minimum angle of resolution chart), 3 years old children were examined using hroken wheel acuity cards or symbols chart. The results of the exminations were conversed and recorded in form of decimal fraction uniformly. ②The corneal reflection and the cover test procedure were used for the position of gaze. ③ Stereoacuity were examined using the stereoptic visual testing cards (made by the Institute of Biophysics, Chinese Academy of Sciences). Stereoacuity were defined as normal when the test value was ≤ 60 s and were defined as abnormal when it was ≥ 100 s. Each examination was performed by same selected physicians. MAIN OUTCOME MEASURFS: The main outcome measurements included ① results of eye position examination and stereoptic examination in children in all age brackets, ② results of optometry in children with strabismus, ③ The relation between eyesight and stereopsis in participants,④Stereoacuity of children in all age brackets, ⑤and possible causations involve in stereopsis. RESULTS: Practically, 8326 participants receiving examinations of eysight and eye position entered the statistical analysis procedure, while an other 662 children were omitted for their poor expression for the examinations. ① Results of stereoptic examination and eye position examination for participants in all age brackets: 165 (2%) with strabismus, in which 89.7%were first diagnosed; 2016 (27.5) with myopia, whose stereoptic value were ≥ 100 s. ②Results of optometry in children with strabismus: 93.5% esotropia occurred in children with hyperopia. The rate of hyperopia/myopia was not significantly different between exotropia and vertical strabismus.③ Relation between eyesight and stereoacuity of participants: 75.5% children have a normal stereoacuity.With a decreasing in eyesight,the stereoacuity decreased (P < 0.05 or P < 0.01). ④ Stereoacuity of the children in all age brackets: With an increasing age, the rate of children with normal stereoacuity increased. ⑤ Possible causations involved in stereopsis: strabismal amblyopia might have the greatest influence on stereopsis. CONCLUSION: ①About 90% children with strabismus were firstly diagnosed in the investigation. Causations responsible for abnormal stereopsis include ametropia, strabismus, and amblyopia, and so on. ② Concomitant esotropia has close relation with binocular accommodation, while exotropia and vertical strabismus are not linked to refractions. Stereoacuity decreases with a decreasing in eyesight. ③ The results of the investigation support the view that mature stereopsis is achieved early than 4 years old although certain variance of stereoptic development can be observed among individuals.

ABSTRACT

@# Presented in this paper is a report on the Quadriplegia Index of Function(QIF) in usewith 16 cases of quadriplegic patients.Compared with the Barthel Index and Japan ADL Scale,QIF wasfound to be more sensitive and effective to document the small but significant funtional gains made byquadriplegics during medical rehabilitation,therefore,authors suggest that QIF should be widely usedin the rehabilitation evaluation of quadriplegic patients.
